Isolation of Cholesterol Sulfate from Bovine Adrenals

the spectra illustrated in Fig. 1 confirm the identity of the isolated material. The salt was hydrolyzed by dissolving the sample in 20 ml of tetrahydrofuran containing perchloric acid (5). After 3 hours at 50”, the solution was brought to neutrality with concentrated NH*OH and the tetrahydrofuran was evaporated under vacuum at 45”. The residue was then partitioned between water and ether, and the ether-soluble material, after evaporation of the solvent, was chromatographed on alumina. The radioactive cholesterol, eluted with benzene, was crystallized from a mixture of acetone and methanol. It melted at 1422145” and when mixed with authentic cholesterol, the mixture melted at 145-148”. Its infrared spectrum was identical in every respect with that of authentic cholesterol. Based on the recovered radioactivity it appears that bovine adrenals contain at least 1.5 mg of cholesterol sulfate per kg. The implications inherent in the observation that sulfated steroidal intermediat,es are involved in hormone formation have been discussed previously (l-3). Now that cholesterol sulfate has been shown to occur in a mammalian tissue the possibility must also be considered that this conjugate may represent an active form of cholesterol in metabolic processes other than those concerned with steroid hormone biosynthesis.
